Influence of phenolic compounds on the growth and arginine deiminase system in a wine lactic acid bacterium BJM
Alberto,María R.; Manca de Nadra,María C.; Arena,Mario E..
The influence of seven phenolic compounds, normally present in wine, on the growth and arginine deiminase system (ADI) of Lactobacillus hilgardii X1B, a wine lactic acid bacterium, was established. This system provides energy for bacterial growth and produces citrulline that reacts with ethanol forming the carcinogen ethyl carbamate (EC), found in some wines. The influence of phenolic compounds on bacterial growth was compound dependent. Growth and final pH values increased in presence of arginine. Arginine consumption decreased in presence of protocatechuic and gallic acids (31 and 17%, respectively) and increased in presence of quercetin, rutin, catechin and the caffeic and vanillic phenolic acids (between 10 and 13%, respectively). ADI enzyme activities...

Simple and robust DNA extraction method for the large-scale analysis of genotypes containing high polyphenolic content, such as landracesof Solanum tuberosum and Zea mays Ciencia e Investigación Agraria
Barra,Macarena; Salazar,Erika; Beltrán,María; Sagredo,Boris.
The extraction of high-quality DNA is essential for studies conducted at the molecular level in species with an abundance of contaminants in their tissues, such as some landraces of potato and maize, in which it is difficult to extract good-quality genomic DNA. Compounds such as polyphenols interfere with the amplification of DNA during polymerase chain reaction (PCR), which makes it difficult to conduct PCR-based studies ofmolecular markers. This article describes a simple and robust protocol for DNA isolation that was applied to potato and maize landraces and resulted in a high DNA concentration with excellent purity and quality. This method is based on the method of Krizman et al. (2006) with some modifications and was tested on potato leaves and young...

A Pyranine based Procedure for Evaluation of the Total Antioxidant Potential (TRAP) of Polyphenols: A Comparison with closely related Methodologies Biol. Res.
CAMPOS,ANA MARÍA; SOTOMAYOR,CARLOS PATRICIO; PINO,EDUARDO; LISSI,EDUARDO.
A novel procedure for the evaluation of total reactive antioxidant potentials (TRAP) is described. The method is based on the measurement of the bleaching of pyranine by peroxyl radicals. The addition of the antioxidants produces a clear induction time whose magnitude is directly related to the antioxidant concentration. A comparison of the values obtained with those reported employing closely related methodologies shows that the results are significantly affected by the substrate employed to monitor the steady state free radical concentration. Possible sources of this dependence are discussed

Antidiarrheal effect of extract from the bark of Combretum leprosum in mice Anais da ABC (AABC)
CAVALCANTI,PAULO M.S.; MARTINS,MARIA DO CARMO C.; NUNES,PAULO H.M.; ALVES FILHO,FRANCISCO C.; SILVA,JANYERSON D.P.; CAVALCANTI,SUZANA M.G..
Abstract This study investigated the effects of the ethanolic extract from the bark of Combretum leprosum (ECL) on intestinal transit and castor-oil induced diarrhea in mice. The oral administration of ECL (750 and 1000 mg/kg) slowed intestinal transit (ID50 of 455 mg/kg). The ECL (250-1000 mg/kg) reduced castor-oil induced diarrhea, in a time- and dose-dependent manner (p < 0.05). To determine if antidiarrheal effect of ECL involves α2-adrenergic or opioid receptor activation, the mice were pretreated with antagonists of these receptors, yohimbine or naloxone respectively. None of these drugs inhibited the antidiarrheal effect of ECL. To test if antidiarrheal effect of ECL is due to an antisecretory action, we realized the enteropooling assay on rats....

Isolation of high-quality RNA from Platycladus orientalis and other Cupressaceae plants Electron. J. Biotechnol.
Chang,Ermei; Zhao,Yuxin; Wei,Qianchun; Shi,Shengqing; Jiang,Zeping.
Platycladus orientalis has a lifespan of several thousand years in China, making it a good plant in which to study aging at the molecular level, but this requires sufficient quantities of high-quality P. orientalis RNA. However, no appropriate methods have been reported for total RNA isolation from P. orientalis leaves. The TRIzol method did not extract RNA, while cetyltrimethylammonium bromide, sodium dodecyl sulfate-phenol, and plant RNAout kit (Tianz, Inc., China) protocols resulted in low yields of poor quality RNA. Isolating total RNA using the Spectrum™ Plant Total RNA Kit (Sigma-Aldrich, St. Louis, MO, USA) resulted in a high-quality product but a low yield. However, the two-step removal of polyphenols and polysaccharides in the improved plant...

Scale-up in the synthesis of nanoparticles for encapsulation of agroindustrial active principles Ciência e Agrotecnologia
Duarte,Dirliane Santos; Nascimento,José Augusto de Almeida; Britto,Douglas de.
ABSTRACT Given the potentiality of nanoparticles (NP) to load substances as active principle of drugs, cosmetics and food, there is interest in increasing its production. Particularly in the agroindustrial area, the byproducts are source of functional compounds that must be stabilized by encapsulation, for example, to allow its application. Considering the amount of generated byproducts, it is necessary to understand the encapsulation synthesis in a high scale dimension. In this way, the active compounds vitamin C (from the byproduct of the acerola processing) and polyphenols (from the byproduct of grape processing) were nanoencapsulated into chitosan NP at three reactional volumes: 0.1; 1.0 and 10.0 dm3. The NP were characterized for yield, particle size,...
